South Africa will play the Bristol Bears at Ashton Gate in November.
The match is part of the fall tour of the world champions in Europe and takes place on Thursday 17 November.
The Springboks will face England, Ireland, France and Italy, with South Africa’s 15th meeting with Bristol two days before they test with Italy.
“This is a huge opportunity for all of our players to test themselves against one of the best players,” Bristol rugby club manager Pat Lamm told the club’s website.
Bristol last played internationally in 2003 when the United States visited Memorial Stadium.
Chairman Chris Bowie added: “This is an important achievement for the club and really underscores how far we have come like bears.”
“Hosting the world champions in our home is very exciting and it will be an event that will go down in history.
“South Africa is one of the most recognizable international teams in the world, with a vibrant and passionate fan base.
“We are grateful to the South African Rugby Union, Premiership Rugby and World Rugby for their support and cooperation in helping us deliver this unique and historic match.”
